TLC Analysis of Cranberry Juice Powder
Sample:
Dissolve 1 g of cranberry juice powder and 1 g of cranberry raw material in 10 mL of methanol, respectively for 30 minute.
After standing for 5 minutes,the mixture is filtered.
The filter is washed with 1 mL methanol.
Solutions are combined and evaporated to dryness.
The residue is dissolved in 1 mL methanol.
Chromatographic conditions:
Stationary phase: TLC plates silica gel 60 F254, 10x10 cm or 20x10 cm.
Mobile Phase: Butyl alcohol: Acetic acid: Water
(4:1:5 V/V/V developed with the upper solution)
Sample application: 2 μL or 5 µL test solution are applied as 100 mm bands, 30 min. 10 mm apart, 100 mm from lower edge of plate.
Development: 10x10 cm or 20x10 cm Twin Trough Chamber, saturated for 20 min (filter paper), 5 mL (respectively 10 mL) developing solvent per trough developing distance 100 mm from lower edge of plate. The plate is then dried with a hair dryer (warm air) for 5 min.
Detection: Examination in natural light and 365nm.
